Test Order Name - Mpox virus PCR

Methodology - Real-time polymerase chain reaction (PCR)

Acceptable Specimen Type(s) for Testing - Lesion swabs collected in Viral Transport Medium (VTM) or Universal Viral Transport media (UVT)

Transport / Collection Medium -

  • Follow personal protection equipment guidance when collecting specimen.
  • Mpox lesions may appear in different stages on different parts of the body. Sample two (2) mpox (MPX) lesions per patient preferably from different body sites. Each lesion should be sampled with one (1) swab.
  • Please submit specimens collected in Universal Viral Transport media (UVT) or Viral transport media (VTM).
  • Vigorously swab lesion with the swab enclosed in the VTM or UVT collection kit. Lesion may unroof, but this is not required. Be aware that swabbing may cause pain to the patient; pressure is necessary for adequate sample collection.
  • Place each swab into separate VTM or UVT collection tubes and break swab at the score mark. DO NOT FORCE the swab into the transport container.
  • Sample, collect, label, and store each specimen in separate biohazard bags. Place the printed requisition on the sleeve outside of the biohazard bag.

Storage and Preservation of Specimen -

  • Store all specimens at 4°C if shipping within 24 to 72 hours; store at -80C if shipping will be delayed.
  • Ship specimens on dry ice in compliance with DoT and IATA regulations governing Biological Substances, Category B (UN 3373) Monday through Thursday. Specimen can also be sent via courier Monday through Friday 8:00am to 5:00pm.

Minimum Volume Required - Not applicable

Additional Collection Instructions - Requisition forms are required for all specimens

  • Please place one (1) order for each lesion being swabbed.
  • SF Public Health Lab will not accept more than 2 lesions per patient.
  • Please email completed CMR to monkeypox@sfdph.org

Additional Required Information -

  • Collection date
  • Clearly marked specimen source, e.g., “L elbow swab” and “R Cheek swab”.
  • Patient’s name and date of birth

Send Out? - No

Turnaround Time - Specimen tested 3 times a week: Monday, Wednesday and Friday.

Testing Restrictions - None
